Overview

Ubigene gene knockout cell lines mediated by optimized CRISPR-Cas9 technology provide reliable in vitro models for investigating gene function, signaling pathways, disease mechanisms, and therapeutic targets.

COP1 (COP1 E3 ubiquitin ligase) encodes an E3 ubiquitin ligase that participates in ubiquitin-dependent protein degradation. As a component of the CUL4A-RING E3 ubiquitin ligase complex, COP1 promotes ubiquitination of target proteins and their subsequent proteasomal turnover. It contributes to regulation of protein homeostasis and cellular responses to stressors including ionizing radiation.

Human cardiac ventricular cells are represented by AC16, an immortalized cell line derived from primary human ventricular tissue. The cells were generated by fusion of primary adult ventricular cardiomyocytes with a human fibroblast-like cell line carrying a temperature-sensitive SV40 large T antigen. AC16 exhibits cardiac-related characteristics and is used as an in vitro model for studies of human cardiomyocyte biology.

Each knockout cell line is validated by STR authentication, sterility testing (bacteria/fungi), and mycoplasma screening. Genotype is confirmed by two rounds of PCR and Sanger sequencing. Only clones that pass all QC criteria are released, ensuring reliable identity and performance for downstream applications.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the COP1 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into AC16 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, COP1 Knockout cell line (AC16) is expanded and cryopreserved for downstream applications.
