Overview

Ubigene gene knockout cell lines mediated by optimized CRISPR-Cas9 technology provide reliable in vitro models for investigating gene function, signaling pathways, disease mechanisms, and therapeutic targets.

Epha2 (Eph receptor A2) encodes a receptor tyrosine kinase in the Eph family, which mediates cell-to-cell signaling involved in tissue organization and development. EPHA2 is associated with processes including blood vessel morphogenesis, nervous system development, lens fiber cell morphogenesis, mammary gland development, and osteoclast differentiation. The protein localizes to the cell surface and is expressed across multiple tissues. Mutations in its human ortholog are associated with several forms of cataract.

The MC3T3-E1 cell line is a foundational mouse osteoblast model derived from the cranial calvaria of newborn C57BL/6 mice. As referenced in image_2ede08.png, a series of subclones were isolated from the cloned but phenotypically heterogeneous parental MC3T3-E1 line to select for varying degrees of osteoblast differentiation and mineralization potential. This adherent line serves as a gold-standard in vitro platform for studying bone morphogenesis, osteogenesis pathway kinetics, and transcriptional regulation during matrix calcification.

Each knockout cell line is validated by STR authentication, sterility testing (bacteria/fungi), and mycoplasma screening. Genotype is confirmed by two rounds of PCR and Sanger sequencing. Only clones that pass all QC criteria are released, ensuring reliable identity and performance for downstream applications.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the Egfr gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into MC3T3-E1 Subclone 14 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, Egfr Knockout cell line (MC3T3-E1 Subclone 14) is expanded and cryopreserved for downstream applications.

Key Advantages

Easy to use
Easy to use

Cas9 cell lines in our cell bank can stably express Cas9 protein. Each Cas9 Stable Cell Line is easy to use and enables gene knockout simply by transfecting gRNA, while transfection of gRNA and donor DNA results in gene knock-in or point mutations

High-efficient KO
High-efficient KO

These Cas9 stable cell lines have been used to construct the KO cell lines for various genes, gene KO efficiency 5-10 times improved.

Curated cells
Curated cells

Selected cells, low passages, good cell condition, high activity, applicable for all kinds of gene-editing experiments.
