Overview

Gene knockout cell lines are generated using CRISPR-Cas9 based technology to precisely disrupt target genes, enabling functional genomics research, disease modeling, and target validation.

4T1 is a murine mammary carcinoma cell line selected as a 6-thioguanine-resistant variant from the 410.4 tumor without mutagen treatment. The cells are used as an aggressive mouse breast cancer model and are capable of spontaneous metastasis in syngeneic BALB/c mice. 4T1 is widely applied in studies of breast cancer progression, tumor metastasis, immune responses, and evaluation of anticancer therapies, particularly in immunocompetent mouse models.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the Fabp5 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into 4T1 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, Fabp5 Knockout cell line (4T1) is expanded and cryopreserved for downstream applications.
