Overview

Ubigene gene knockout cell lines mediated by optimized CRISPR-Cas9 technology provide reliable in vitro models for investigating gene function, signaling pathways, disease mechanisms, and therapeutic targets.

A human liver tumor cell line established from the tumor tissue of a 15-year-old Caucasian male in 1975. Originally described as a well-differentiated hepatocellular carcinoma, it has subsequently been reclassified by some sources as a hepatoblastoma. The cells produce multiple plasma proteins, including albumin, alpha-fetoprotein, alpha2-macroglobulin, alpha1-antitrypsin, transferrin, plasminogen, and complement components, and have been reported to respond to stimulation with human growth hormone.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the GFI1B gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into Hep G2 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, GFI1B Knockout Cell line (Hep G2) is expanded and cryopreserved for downstream applications.
