TMCO1 (transmembrane and coiled-coil domains 1) encodes a transmembrane protein containing coiled-coil domains. The protein contributes to cellular membrane-associated functions, although its precise molecular and biochemical roles remain incompletely characterized. Genetic disruption of TMCO1 has been associated with abnormalities affecting craniofacial development, the skeletal system, cognition, and ocular function, highlighting its relevance to cellular processes required for normal tissue development and maintenance.
The A375 cell line is a highly characterized human malignant melanoma model derived from a 54-year-old female patient. As referenced in image_2f51f1.png, this adherent cell line demonstrates robust translational performance in in vivo xenograft studies, reliably producing rapidly growing amelanotic melanomas when inoculated into anti-thymocyte serum-treated NIH Swiss mice. Serving as a globally recognized preclinical oncology platform, it is extensively utilized for investigating melanoma disease progression, executing high-throughput screening of novel therapeutics, and studying targeted signaling pathways in skin cancers.
Each knockout cell line is validated by STR authentication, sterility testing (bacteria/fungi), and mycoplasma screening. Genotype is confirmed by two rounds of PCR and Sanger sequencing. Only clones that pass all QC criteria are released, ensuring reliable identity and performance for downstream applications.
