Overview

Ubigene gene knockout cell lines mediated by optimized CRISPR-Cas9 technology provide reliable in vitro models for investigating gene function, signaling pathways, disease mechanisms, and therapeutic targets.

Tpm1 (tropomyosin 1) encodes a member of the tropomyosin protein family that forms heterodimers with a beta subunit and binds actin filaments in muscle and nonmuscle cells. In smooth and striated muscle, TPM1 contributes to calcium-dependent regulation of muscle contraction. In nonmuscle cells, it helps stabilize the actin cytoskeleton. Alternative splicing generates multiple transcript variants encoding distinct protein isoforms, supporting diverse functions across different cellular contexts.

MPC-5 is a mouse plasma cell tumor-derived cell line established from the Merwin plasma cell tumor-II carried in BALB/c mice. The cells retain plasma cell characteristics and produce fully assembled immunoglobulin G2b (IgG2b), as well as immunoglobulin-related subunits including light chain dimers and free light chains. MPC-5 is used as a model for studying plasma cell biology, immunoglobulin production, and antibody synthesis mechanisms.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the Tollip gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into MPC-5 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, Tollip Knockout cell line (MPC-5) is expanded and cryopreserved for downstream applications.

Product Overview

Tollip Knockout cell line (MPC-5)
The Tollip Knockout cell line (MPC-5) is generated by targeted disruption of the Tollip gene in MPC-5 cells. This cell line is designed to investigate the role of Tollip in cellular function and gene regulation, as well as its potential involvement in disease-related pathways.

Key Advantages

Easy to use
Easy to use

Cas9 cell lines in our cell bank can stably express Cas9 protein. Each Cas9 Stable Cell Line is easy to use and enables gene knockout simply by transfecting gRNA, while transfection of gRNA and donor DNA results in gene knock-in or point mutations

High-efficient KO
High-efficient KO

These Cas9 stable cell lines have been used to construct the KO cell lines for various genes, gene KO efficiency 5-10 times improved.

Curated cells
Curated cells

Selected cells, low passages, good cell condition, high activity, applicable for all kinds of gene-editing experiments.
