Overview

CRISPR knockout cell lines are engineered through targeted genome editing to eliminate gene function, supporting studies of cellular biology, disease mechanisms, and drug discovery.

ZC3HC1 (zinc finger C3HC-type containing 1) encodes an F-box-containing protein that functions as a component of an SCF-type E3 ubiquitin ligase complex. The complex regulates cell-cycle progression by targeting cyclin B1 for ubiquitin-mediated degradation, thereby controlling the transition from G2 phase to mitosis. ZC3HC1 therefore contributes to regulation of cell-cycle timing through targeted protein degradation.

A human hepatocellular carcinoma cell line of Japanese origin, established from the liver tumor of a 57-year-old male patient. The cells are classified as an adult hepatocellular carcinoma line and have been extensively characterized for their molecular and genetic properties. The cell line exhibits a predominantly East Asian genomic ancestry profile and carries documented sequence variations affecting genes including KDR, POLD3, TERT, and TP53.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZC3HC1 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into HuH-7 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZC3HC1 Knockout cell line (HuH-7) is expanded and cryopreserved for downstream applications.
