Overview

Ubigene gene knockout cell lines mediated by optimized CRISPR-Cas9 technology provide reliable in vitro models for investigating gene function, signaling pathways, disease mechanisms, and therapeutic targets.

ZC3HC1 (zinc finger C3HC-type containing 1) encodes an F-box-containing protein that functions as a component of an SCF-type E3 ubiquitin ligase complex. The complex regulates cell-cycle progression by targeting cyclin B1 for ubiquitin-mediated degradation, thereby controlling the transition from G2 phase to mitosis. ZC3HC1 therefore contributes to regulation of cell-cycle timing through targeted protein degradation.

LoVo is a human colorectal adenocarcinoma cell line established from a metastatic tumor in the left supraclavicular region of a 56-year-old male patient. The cells produce carcinoembryonic antigen (CEA) and represent a metastatic colorectal cancer model. LoVo is used for studies of colorectal cancer biology, tumor metastasis, and cancer-related signaling mechanisms.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZC3HC1 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into LoVo cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZC3HC1 Knockout cell line (LoVo) is expanded and cryopreserved for downstream applications.
