ZDHHC12 (zDHHC palmitoyltransferase 12) encodes a protein-cysteine S-palmitoyltransferase involved in protein palmitoylation. The protein localizes primarily to the Golgi apparatus and endoplasmic reticulum and contributes to gephyrin clustering associated with postsynaptic density assembly. ZDHHC12 also participates in negative regulation of NLRP3 inflammasome complex assembly, linking protein lipid modification with neuronal and inflammatory cellular processes.

A human esophageal cancer cell line established from a poorly differentiated squamous cell carcinoma of the upper cervical esophagus in a 49-year-old woman after radiotherapy. KYSE-150 carries amplified c-erb-B and cyclin D1 oncogenes and has been reported to produce tumors in nude mice. The cell line has been extensively characterized and is included in large-scale cancer cell line resources.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZDHHC12 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into KYSE-150 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZDHHC12 Knockout cell line (KYSE-150) is expanded and cryopreserved for downstream applications.
