Gene knockout cell lines are generated using CRISPR-Cas9 based technology to precisely disrupt target genes, enabling functional genomics research, disease modeling, and target validation.

Zfp36 (zinc finger protein 36) encodes an RNA-binding protein that recognizes AU-rich regions in the 3′ untranslated regions of messenger RNAs. It participates in post-transcriptional gene regulation and is associated with the CCR4-NOT and ribonucleoprotein complexes. ZFP36 regulates RNA metabolism and signaling processes, including the p38 MAPK cascade, and contributes to the control of cellular differentiation. It is expressed across multiple anatomical systems and has been studied in relation to rheumatoid arthritis. Its human ortholog is ZFP36.

VERO C1008 (E6) is a subclone of the Vero76 cell line, which was originally derived from the kidney of an African green monkey. The cells retain susceptibility to a range of viruses and exhibit some degree of contact inhibition, making them suitable for supporting the growth and study of slowly replicating viruses. VERO C1008 (E6) is widely used in virology research, including viral propagation, infection studies, and evaluation of viral replication.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZDHHC20 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into VERO C1008 (E6) cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZDHHC20 Knockout cell line (VERO C1008 (E6)) is expanded and cryopreserved for downstream applications.
