ZDHHC9 (zDHHC palmitoyltransferase 9) encodes an integral membrane palmitoyltransferase belonging to the zinc finger DHHC-domain family. The protein forms a complex with GOLGA7 and catalyzes palmitoylation of Ras family proteins including HRAS and NRAS. ZDHHC9 is relevant to studies of protein lipid modification, membrane-associated signaling, and regulation of Ras protein activity.

AGS is a human gastric adenocarcinoma cell line established from a primary stomach tumor of a 54-year-old female patient who had not received prior anticancer treatment. The cells are derived from gastric adenocarcinoma and are used as a model for studying gastric cancer biology, tumor-related signaling pathways, infection-related mechanisms, and therapeutic responses. AGS cells have been reported to carry persistent parainfluenza virus type 5 (PIV5) infection.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZDHHC9 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into AGS cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZDHHC9 Knockout cell line (AGS) is expanded and cryopreserved for downstream applications.
