Overview

CRISPR knockout cell lines are engineered through targeted genome editing to eliminate gene function, supporting studies of cellular biology, disease mechanisms, and drug discovery.

ZFAND5 (zinc finger AN1-type containing 5) encodes a zinc finger-containing protein with predicted DNA-binding and zinc ion-binding activities. It is predicted to participate in regulation of processes including fibroblast migration and platelet-derived growth factor receptor signaling and is localized primarily to the cytoplasm. ZFAND5 is relevant to studies of zinc finger protein function, cellular signaling, cell migration, and regulation of gene expression.

A human urinary bladder transitional papillary tumor cell line established in 1968 from a recurrent, well-differentiated papillary tumor of a 63-year-old man. The original tumor was classified as clinical stage T2 and histological grade G1 and had been treated previously with gold grains and subsequently with diathermy following recurrence.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZFAND5 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into RT4 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZFAND5 Knockout cell line (RT4) is expanded and cryopreserved for downstream applications.
