Overview

CRISPR knockout cell lines are engineered through targeted genome editing to eliminate gene function, supporting studies of cellular biology, disease mechanisms, and drug discovery.

ZFP36 (ZFP36 zinc finger CCCH-type) encodes an RNA-binding regulatory protein that recognizes AU-rich regions in the 3′ untranslated regions of target mRNAs. It participates in regulation of gene expression and mRNA catabolism and responds to cytokine and epidermal growth factor signaling. Through sequence-specific RNA interactions, ZFP36 contributes to post-transcriptional control of gene expression and cellular responses to extracellular stimuli.

LoVo is a human colorectal adenocarcinoma cell line established from a metastatic tumor in the left supraclavicular region of a 56-year-old male patient. The cells produce carcinoembryonic antigen (CEA) and represent a metastatic colorectal cancer model. LoVo is used for studies of colorectal cancer biology, tumor metastasis, and cancer-related signaling mechanisms.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZFP36 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into LoVo cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZFP36 Knockout cell line (LoVo) is expanded and cryopreserved for downstream applications.
