ZFPL1 (Zinc finger protein like 1) encodes a zinc-binding protein predicted to function in DNA binding and transcriptional regulation. ZFPL1 is localized to the Golgi apparatus and may contribute to regulation of gene expression through interactions involving zinc finger domains.

The K-562 cell line was established by Lozzio and Lozzio in 1970 from the pleural effusion of a 53-year-old female patient with chronic myeloid leukemia (CML) in blast crisis. The cells carry the BCR-ABL1 e14-a2 (b3-a2) fusion gene and exhibit characteristics of myeloid leukemia cells, including hemoglobin production. K-562 is a human leukemia cell model widely used for studying CML biology, myeloid differentiation, hematopoietic signaling, and immune cell interactions, including applications as an in vitro target model in natural killer (NK) cell assays.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZFPL1 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into K-562 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZFPL1 Knockout cell line (K-562) is expanded and cryopreserved for downstream applications.
