5637 is a human bladder carcinoma cell line established from a primary bladder tumor of a 68-year-old male patient. The cells are known to produce multiple growth factors, including stem cell factor (SCF), interleukins, and colony-stimulating factors. 5637 cells are used as a model for studying bladder cancer biology, tumor-associated signaling, and growth factor regulation.

Each knockout cell line is validated by STR authentication, sterility testing (bacteria/fungi), and mycoplasma screening. Genotype is confirmed by two rounds of PCR and Sanger sequencing. Only clones that pass all QC criteria are released, ensuring reliable identity and performance for downstream applications.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZNF185 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into 5637 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZNF185 Knockout cell line (5637) is expanded and cryopreserved for downstream applications.
