ZNF32 (zinc finger protein 32) encodes a zinc finger protein that enables sequence-specific binding to double-stranded DNA. It is predicted to function in the regulation of transcription by RNA polymerase II and is primarily associated with the nucleus. Its DNA-binding activity suggests a role in transcriptional regulation and chromatin-associated processes, making ZNF32 relevant for studying mechanisms that control gene expression and transcriptional activity.

Papillary thyroid carcinoma is represented by TPC-1, a human thyroid cancer cell line established from a thyroid tumor. The cells are classified as a human papillary thyroid carcinoma model and carry the characteristic RET/PTC1 rearrangement resulting from fusion of RET with CCDC6. TPC-1 is widely used for studies of thyroid cancer biology and RET-associated oncogenic signaling.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZNF32 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into TPC-1 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZNF32 Knockout cell line (TPC-1) is expanded and cryopreserved for downstream applications.
