ZNF354A (zinc finger protein 354A) encodes a predicted zinc finger transcription factor capable of sequence-specific DNA binding at RNA polymerase II regulatory regions. The protein is predicted to participate in regulation of RNA polymerase II-dependent transcription and is associated with nuclear and cytoplasmic compartments. Its functional characterization remains limited, but its predicted DNA-binding activity supports a role in transcriptional regulation.

LoVo is a human colorectal adenocarcinoma cell line established from a metastatic tumor in the left supraclavicular region of a 56-year-old male patient. The cells produce carcinoembryonic antigen (CEA) and represent a metastatic colorectal cancer model. LoVo is used for studies of colorectal cancer biology, tumor metastasis, and cancer-related signaling mechanisms.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZNF354A gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into LoVo cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZNF354A Knockout cell line (LoVo) is expanded and cryopreserved for downstream applications.
