ZNF432 (zinc finger protein 432) encodes a zinc finger protein with damaged DNA-binding and protein-macromolecule adaptor activities. It participates in the cellular DNA damage response and is localized in the nucleoplasm. Through its DNA- and protein-binding properties, ZNF432 may contribute to coordination of molecular responses to damaged DNA, although its specific targets and mechanisms remain incompletely characterized.

A human T-cell line originating from the peripheral blood of a 14-year-old male with childhood T-cell acute lymphoblastic leukemia, Jurkat, Clone E6-1 represents a clone of the Jurkat-FHCRC derivative. The line is classified as a T-cell lymphoblast and has been characterized with genomic and proteomic studies. It is associated with precursor T-cell acute lymphoblastic leukemia and retains the biological characteristics of its leukemia-derived origin.

Before delivery, each clone is authenticated by STR, tested for sterility and mycoplasma, and genotype‑verified by two‑round PCR‑sequencing. Comprehensive QC data accompany every shipment to support reproducible research.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZNF432 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into Jurkat, Clone E6-1 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZNF432 Knockout cell line (Jurkat, Clone E6-1) is expanded and cryopreserved for downstream applications.
