Overview

Gene knockout cell lines are generated using CRISPR-Cas9 based technology to precisely disrupt target genes, enabling functional genomics research, disease modeling, and target validation.

ZNHIT1 (zinc finger HIT-type containing 1) encodes a nuclear protein with histone-binding activity. It is involved in muscle cell differentiation and contributes to positive regulation of DNA damage responses mediated by p53-class signaling. Through its interactions with histones and participation in cellular response pathways, ZNHIT1 may contribute to coordination of chromatin-associated regulation, differentiation, and genome damage responses.

LoVo is a human colorectal adenocarcinoma cell line established from a metastatic tumor in the left supraclavicular region of a 56-year-old male patient. The cells produce carcinoembryonic antigen (CEA) and represent a metastatic colorectal cancer model. LoVo is used for studies of colorectal cancer biology, tumor metastasis, and cancer-related signaling mechanisms.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZNHIT1 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into LoVo cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZNHIT1 Knockout cell line (LoVo) is expanded and cryopreserved for downstream applications.
