Overview

CRISPR knockout cell lines are engineered through targeted genome editing to eliminate gene function, supporting studies of cellular biology, disease mechanisms, and drug discovery.

ZP3 (zona pellucida glycoprotein 3) encodes a structural glycoprotein of the zona pellucida surrounding mammalian oocytes and early embryos. ZP3 participates in sperm binding and induction of the sperm acrosome reaction, making it an important component of fertilization. The protein contains a conserved zona pellucida domain and is processed to form the mature extracellular matrix component. Its functions are closely associated with fertilization and preimplantation development.

NCI-H1975 is a human non-small cell lung cancer cell line established from lung tumor tissue. The donor was a non-smoker. The cell line is used as a model for studying lung cancer biology and molecular mechanisms associated with non-small cell lung carcinoma.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy

Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the ZP3 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into NCI-H1975 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, ZP3 Knockout cell line (NCI-H1975) is expanded and cryopreserved for downstream applications.
