采用独家红棉系统（www.rc-crispr.com）设计Gpx4基因的sgRNA片段敲除方案，将两条sgRNA构建到CRISPR-U™载体上，通过电转法/病毒法将载体转染进MDCK(NBL-2)细胞中，使用极限稀释法制备单克隆，并利用EZ-editor™单克隆基因型鉴定试剂盒（货号：YK-MV-1000）对单克隆进行核酸裂解及PCR扩增，再通过Sanger 测序验证基因型，复检合格后对Gpx4基因敲除MDCK(NBL-2)细胞进行扩增和冻存。
