Overview

CRISPR knockout cell lines are engineered through targeted genome editing to eliminate gene function, supporting studies of cellular biology, disease mechanisms, and drug discovery.

AAGAB (alpha and gamma adaptin binding protein) encodes a protein that interacts with alpha- and gamma-adaptin subunits of complexes involved in clathrin-coated vesicle trafficking. Through these interactions, AAGAB contributes to intracellular membrane trafficking and clathrin-dependent vesicle transport. Its function is relevant to studies of endocytic pathways and regulation of cellular membrane dynamics.

A human T-cell line originating from the peripheral blood of a 14-year-old male with childhood T-cell acute lymphoblastic leukemia, Jurkat, Clone E6-1 represents a clone of the Jurkat-FHCRC derivative. The line is classified as a T-cell lymphoblast and has been characterized with genomic and proteomic studies. It is associated with precursor T-cell acute lymphoblastic leukemia and retains the biological characteristics of its leukemia-derived origin.

Each knockout cell line is validated by STR authentication, sterility testing (bacteria/fungi), and mycoplasma screening. Genotype is confirmed by two rounds of PCR and Sanger sequencing. Only clones that pass all QC criteria are released, ensuring reliable identity and performance for downstream applications.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the AAGAB gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into Jurkat, Clone E6-1 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, AAGAB Knockout cell line (Jurkat, Clone E6-1) is expanded and cryopreserved for downstream applications.
