Overview

Gene knockout cell lines are generated using CRISPR-Cas9 based technology to precisely disrupt target genes, enabling functional genomics research, disease modeling, and target validation.

The RAW 264.7 cell line was established from the ascites of a tumor induced in a male mouse by intraperitoneal injection of Abelson murine leukemia virus (A-MuLV). This murine macrophage-like cell line exhibits functional characteristics associated with macrophages, including pinocytosis of neutral red, phagocytosis of zymosan particles, and antibody-dependent lysis of target cells. RAW 264.7 cells are responsive to lipopolysaccharide (LPS), which inhibits cell growth, and are widely used as an in vitro model for studying macrophage activation, innate immune responses, inflammatory signaling, and related immunological processes.

Each knockout cell line is validated by STR authentication, sterility testing (bacteria/fungi), and mycoplasma screening. Genotype is confirmed by two rounds of PCR and Sanger sequencing. Only clones that pass all QC criteria are released, ensuring reliable identity and performance for downstream applications.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the Irak3 gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into RAW 264.7 cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, Irak3 Knockout cell line (RAW 264.7) is expanded and cryopreserved for downstream applications.
