Overview

Ubigene gene knockout cell lines mediated by optimized CRISPR-Cas9 technology provide reliable in vitro models for investigating gene function, signaling pathways, disease mechanisms, and therapeutic targets.

MYL6B (myosin light chain 6B) encodes an alkali myosin light chain expressed in skeletal muscle and non-muscle tissues. The protein contributes to regulation of myosin motor complex function and actomyosin-based cellular processes.

JAR is a human choriocarcinoma cell line established from a trophoblastic tumor of the placenta of a 24-year-old woman. The cell line originates from a male fetus and represents a trophoblast-derived tumor model. JAR is used for studies of placental biology, trophoblast differentiation, and mechanisms involved in gestational cancers.

Comprehensive quality control includes STR authentication, sterility testing (bacteria and fungi), and mycoplasma screening to ensure purity and performance. Genotype is confirmed by two rounds of PCR and Sanger sequencing, and complete validation reports are provided for make-to-order products.

KO Cell Construction Strategy
Adopting Red Cotton CRISPR Gene Editing Designer (www.rc-crispr.com), two sgRNAs targeting the MYL6B gene are designed to generate a knockout strategy. The two sgRNA sequences are subsequently cloned into the EZ-editor™ vector and introduced into JAR cells via electroporation or lentiviral transduction. Single-cell clones are then generated using the limiting dilution method. Genomic DNA from individual clones is subjected to nucleic acid lysis and PCR amplification using the EZ-editor™ Monoclone Genotype Validation Kit (Cat# YK-MV-1000). The edited loci are further verified by Sanger sequencing to confirm the genotype. After secondary validation and quality confirmation, MYL6B Knockout cell line (JAR) is expanded and cryopreserved for downstream applications.
